Changes of Differential Urinary Metabolites after High-Intensive Training in Teenage Football Players.

Abstract
OBJECTIVE The mechanism underlying the fatigue of football players is closely related to the energy depletion and accumulation of metabolites; the present study tries to explore the metabolic mechanism in teenage football players during exercise-induced fatigue. METHODS 12 teenage football players were subjected to three groups of combined training by using a cycle ergometer, with the subjective Rating of Perceived Exertion (RPE) as a fatigue criterion. The following indicators were measured in each group after training: maximum oxygen uptake (VO2max), anaerobic power, and average anaerobic power. Urine samples were collected before and after the training. Gas chromatography-mass spectrometry (GC-MS) was performed for the metabonomics analysis of the samples. The metabolism data was analyzed by using principal component analysis (PCA) and orthogonal partial least squares analysis (OPLS-DA), through the Kyoto Encyclopedia of Genes and Genomes (KEGG) database to confirm the potential differences between metabolites, and the MetPA database was used to analyze the related metabolic pathways. RESULTS There was no significant difference between the maximal oxygen uptakes among the three groups. Compared with group 1, the maximum and average anaerobic power in group 3 significantly decreased (p < 0.05) at the end of training. GC-MS detected 635 metabolites in the urine samples. Through PCA, OPLS-DA analysis, and KEGG matching, 25 different metabolites (3↑22↓) that met the conditions were finally selected. These different metabolites belonged to 5 metabolic pathways: glycine-serine-threonine metabolism, citrate cycle, tyrosine metabolism, nitrogen metabolism, and glycerophospholipid metabolism. CONCLUSIONS During the combined exercise of aerobic and anaerobic metabolism, teenage football players show a significant decrease in anaerobic capacity after fatigue. The metabolic mechanism of exercise fatigue was related to disorders in amino acid and energy metabolism.

Abstract
This pilot work examined associations of brain grey matter volumes (GMV) with perceived fatigability in older adults to elucidate disablement mechanisms. A subsample (n = 29; age = 77.2 ± 5.5; 86% female) of participants from the Lifestyle Interventions and Independence for Elders (LIFE) Study was utilized to quantify GMV for regions of interest in the basal ganglia and limbic system normalized to intracranial volume. The Pittsburgh Fatigability Scale measured physical and mental fatigability (score 0-50; higher physical fatigability ≥ 15; higher mental fatigability ≥ 13). We used an exploratory alpha level of p < 0.1. Nineteen (66%) participants had higher physical fatigability, 19 (66%) had higher mental fatigability, of these, 17 (57%) had both. Right hippocampal volumes/ICV were smaller in participants with higher verses lower physical fatigability (0.261 ± 0.039 vs. 0.273 ± 0.022, p = 0.07); associations were similar for right putamen and bilateral thalamus. Higher mental fatigability was associated with smaller right hippocampus, thalamus, and posterior cingulum and bilateral amygdala. Higher fatigability in older adults may be associated with smaller volumes of the basal ganglia and limbic system, indicating mechanisms for further exploration.

Abstract
PURPOSE Eastern Cooperative Oncology Group Performance Status (ECOG-PS) is currently an important parameter in the choice of treatment strategy for metastatic pancreatic adenocarcinoma (mPA) patients. However, previous research has shown that patients' self-reported health-related quality of life (HRQOL) scales provided additional prognostic information in homogeneous groups of patients with respect to ECOG-PS. The aim of this study was to identify HRQOL scales with independent prognostic value in mPA and to propose prognostic groups for these patients. METHODS We analysed data from 98 chemotherapy-naive patients with histologically proven mPA recruited from 2007 to 2011 in the FIRGEM phase II study which aimed to compare the effectiveness of two chemotherapy regimen. HRQOL data were assessed with the European Organization for Research and Treatment of Cancer QLQ-C30 questionnaire. A random survival forest methodology was used to impute missing data and to identify major prognostic factors for overall survival. RESULTS Baseline HRQOL assessment was completed by 60 % of patients (59/98). Twelve prognostic variables were identified. The three most important prognostic variables were fatigue, appetite loss, and role functioning, followed by three laboratory variables. The model's discriminative power assessed by Harrell's C statistic was 0.65. Fatigue score explained almost all the survival variability. CONCLUSION HRQOL scores have prognostic value for mPA patients with good ECOG-PS. Moreover, the patient's fatigue, appetite loss, and self-perception of daily activities were more reliable prognostic indicators than clinical and laboratory variables. These HRQOL scores, especially the fatigue symptom, should be urgently included for prognostic assessment of mPA patients (with good ECOG-PS).

Three consecutive days of interval runs to exhaustion affects lymphocyte subset apoptosis and migration.

Abstract
This investigation assessed the lymphocyte subset response to three days of intermittent run exercise to exhaustion. Twelve healthy college-aged males (n = 8) and females (n = 4) (age = 26 ± 4 years; height = 170.2 ± 10 cm; body mass = 75 ± 18 kg) completed an exertion test (maximal running speed and VO2max) and later performed three consecutive days of an intermittent run protocol to exhaustion (30 sec at maximal running speed and 30 sec at half of the maximal running speed). Blood was collected before exercise (PRE) and immediately following the treadmill bout (POST) each day. When the absolute change from baseline was evaluated (i. e., Δ baseline), a significant change in CD4+ and CD8+ for CX3CR1 cells was observed by completion of the third day. Significant changes in both apoptosis and migration were observed following two consecutive days in CD19+ lymphocytes, and the influence of apoptosis persisted following the third day. Given these lymphocyte responses, it is recommended that a rest day be incorporated following two consecutive days of a high-intensity intermittent run program to minimize immune cell modulations and reduce potential susceptibility.

Abstract
The purpose of the present study was to examine oxygen consumption rate ([Vdot][Formula: see text]), heart rate (HR), and ratings of perceived exertion (RPE) responses, as well as time to exhaustion (Tlim) values during continuous rides at critical power (CP) determined from the 3-min all-out test. Eighteen participants (mean ± s: 23.6 ± 3.5 years; 72.7 ± 18.2 kg) performed an incremental cycle ergometer test to exhaustion to determine peak oxygen consumption rate ([Vdot][Formula: see text] peak) and HR peak. Critical power was determined from the 3-min all-out test. Metabolic responses ([Vdot][Formula: see text] and heart rate), RPE, and Tlim were recorded during continuous rides to exhaustion at CP. Linear regression and t-tests were used to compare [Vdot][Formula: see text], heart rate, and RPE responses during the continuous rides to exhaustion. The Tlim at CP was 12.5 ± 6.5 min. There were significant increases in [Vdot][Formula: see text], HR, and RPE during the continuous rides at CP and 15 of the 18 participants reached [Vdot][Formula: see text] peak at exhaustion. Therefore, the [Vdot][Formula: see text], heart rate, and RPE responses, as well as the Tlim values in the present study suggested that CP determined from the 3-min all-out test overestimated the "true" CP and was within the severe exercise intensity domain.

Abstract
An intervention study was carried out to examine the effects of chewing gum on occupational stress and related outcomes. 101 volunteers from Cardiff University completed the study. The results showed that chewing gum reduced stress (both at work and outside work), reduced fatigue, reduced anxiety and depression and led to a more positive mood. Chewing gum was also associated with perceptions of better performance (both at work and outside).

Abstract
PURPOSE This pilot trial sought to investigate whether any of three doses of American ginseng (Panax quinquefolius) might help cancer-related fatigue. A secondary aim was to evaluate toxicity. METHODS Eligible adults with cancer were randomized in a double-blind manner, to receive American ginseng in doses of 750, 1,000, or 2,000 mg/day or placebo given in twice daily dosing over 8 weeks. Outcome measures included the Brief Fatigue Inventory, vitality subscale of the Medical Outcome Scale Short Form-36 (SF-36), and the Global Impression of Benefit Scale at 4 and 8 weeks. RESULTS Two hundred ninety patients were accrued to this trial. Nonsignificant trends for all outcomes were seen in favor of the 1,000- and 2,000-mg/day doses of American ginseng. Area under the curve analysis of activity interference from the Brief Fatigue Inventory was 460-467 in the placebo group and 750 mg/day group versus 480-551 in the 1,000- and 2,000-mg/day arms, respectively. Change from baseline in the vitality subscale of the SF-36 was 7.3-7.8 in the placebo and the 750-mg/day arm, versus 10.5-14.6 in the 1,000- and 2,000-mg/day arms. Over twice as many patients on ginseng perceived a benefit and were satisfied with treatment over those on placebo. There were no significant differences in any measured toxicities between any of the arms. CONCLUSION There appears to be some activity and tolerable toxicity at 1,000-2,000 mg/day doses of American ginseng with regard to cancer-related fatigue. Thus, further study of American ginseng is warranted.

Calcium supplementation and 4-week exercise on blood parameters of athletes at rest and exhaustion.

Abstract
In the present study, experiments were designed to investigate if supplementation with calcium during 4 weeks had an effect on blood parameters in sedentary male athletes at rest and exhaustion. Thirty healthy subjects of ages ranging from 18 to 22 years were included in the study. The subjects were separated into three groups, as follows: Group 1 consisted sedentary athletes receiving 35 mg/kg/day calcium gluconate. Group 2 included subjects equally supplemented with calcium training 90 min/day for 5 days/week. Group 3 were subject to the same exercise regime but did not receive calcium supplements. Blood parameters were determined in the experimental subjects at rest and after exhaustion. The leukocyte count (WBC) of athletes in groups 2 and 3 were significantly higher at exhaustion (p < 0.05). There were no significant differences in the WBC of the two supplemented groups. The erythrocyte count (RBC) was increased in the supplemented athletes after training (p < 0.05), but hemoglobin, hematocrit, and thrombocyte levels remained unchanged. The mean corpuscular volume increased in the calcium-supplemented group at rest (p < 0.05). These results suggest that calcium supplementation only causes increases in white and red blood cell counts in athletes after exhaustion while other hematological parameters remain unchanged.

Carbohydrate availability and muscle energy metabolism during intermittent running.

Abstract
PURPOSE To examine the influence of ingesting a carbohydrate-electrolyte (CHO-E) solution on muscle glycogen use and intermittent running capacity after consumption of a carbohydrate (CHO)-rich diet. METHODS Six male volunteers (mean +/- SD: age 22.7 +/- 3.4 yr; body mass (BM) 75.0 +/- 4.3 kg; V O2 max 60.2 +/- 1.6 mL x kg(-1) x min(-1)) performed two trials separated by 14 d in a randomized, crossover design. Subjects consumed either a 6.4% CHO-E solution or a placebo (PLA) in a double-blind fashion immediately before each trial (8 mL x kg(-1) BM) and at 15-min intervals (3 mL x kg(-1) BM) during intermittent high-intensity running to fatigue performed after CHO loading for 2 d. Muscle biopsy samples were obtained before exercise, after 90 min of exercise, and at fatigue. RESULTS Subjects ran longer in the CHO-E trial (158.0 +/- 28.4 min) compared with the PLA trial (131.0 +/- 19.7 min; P < 0.05). There were no differences in muscle glycogen use for the first 90 min of exercise (approximately 2 mmol of glucosyl units per kilogram of dry matter (DM) per minute). However, there was a trend for a greater use in the PLA trial after 90 min (4.2 +/- 2.8 mmol x kg(-1) DM x min(-1)) compared with the CHO-E trial (2.5 +/- 0.7 mmol x kg(-1) DM x min(-1); P = 0.10). Plasma glucose concentrations were higher at fatigue in the CHO-E than in the PLA trial (P < 0.001). CONCLUSIONS These results suggest that CHO-E ingestion improves endurance capacity during intermittent high-intensity running in subjects with high preexercise muscle glycogen concentrations. The greater endurance capacity cannot be explained solely by differences in muscle glycogen, and it may actually be a consequence of the higher plasma glucose concentration towards the end of exercise that provided a sustained source of CHO for muscle metabolism and for the central nervous system.
