Availability of arsenic in rice grains by in vitro and in vivo (humans) assays.

Abstract
BACKGROUND Rice grains are consumed by approximately half of the world's population. This cereal has higher arsenic (As) concentrations in grains than wheat or barley. Arsenic determination in food and/or in vitro studies are important for risk assessment; however, it is not enough to assess the real human exposure. METHOD In vitro bioaccessibility was carried out in husked-rice using gastric and intestinal solutions similar to humans. Also, As naturally found in husked-rice was evaluated by in vivo bioavailability in humans. For this purpose, diets from the 1st and 2nd days were free of foods known to be high in As; 3rd and 4th days the diets were composed by rice and water and; 5th and 6th the diet was similar the 1st and 2nd days. During all experimentation, a representative aliquot of each meal, blood and urine were collected for total As (t-As) determination. Arsenic species were determined in the urine. RESULTS t-As in husked rice varied from 157.3 ± 30.6 to 240.2 ± 85.2 μg kg-1. The in vitrobioaccessible fractions ranged from 91 to 94%. Inorganic As (i-As) ranged from 99.7 ± 11.2 to 159.5 ± 29.4 μg kg-1. For the in vivo assay, t-As concentrations in the woman and man blood were about 3 μg mL-1 from the 1st to 6th day. Arsenic from the rice ingested was excreted by urine about 72 h after ingestion. The t-As and dimethyl As (DMA) in urine ranged from 3.59 to 47.17 and 1.02 to 2.55 μg g-1 creatinine for the volunteers, indicating a two-fold DMA-increase in urine after ingestion of husked-rice. CONCLUSION After rice ingestion, As was quickly metabolized. The higher As concentrations were found in urine 72 h after rice ingestion. The main As-specie found in urine was DMA, indicating that methylation of As from rice followed by urine excretion is the main biological pathway for As excretion.

Abstract
Vitamin E is a natural lipophilic vitamin, and the most famous function of vitamin E is an antioxidant activity. Because we have α-tocopherol transfer protein, many vitamin E-related reports are about α-tocopherol. Recently, other vitamin E isoforms, tocotrienols are focusing. Because tocotrienols have unique biological functions such as induction of apoptosis, neuroprotective and anti-obesity effects. Tocotrienols contain in annatto, palm, whole wheat and rice bran. Rice is a typical food in the East Asian countries and Japan. Recently, intake of whole rice is a popular in young women of Japan. Previously, we demonstrated that treatment with tocotrienols on the neuronal cells shows a strong antioxidant effect compared to the tocopherols. In this review, I introduce about neuroprotective and anti-obesity effects of tocotrienols. I would like to show daily intake of whole rice is very good for our health in this review.

Abstract
The blast disease, caused by the ascomycete Magnaporthe oryzae, poses a great threat to rice production worldwide. Increasing use of fungicides and/or blast-resistant varieties of rice (Oryza sativa) has proved to be ineffective in long-term control of blast disease under field conditions. To develop effective and durable resistance to blast, it is important to understand the cellular mechanisms underlying pathogenic development in M. oryzae. In this review, we summarize the latest research in phototropism, autophagy, nutrient and redox signaling, and intrinsic phytohormone mimics in M. oryzae for cellular and metabolic adaptation(s) during its interactions with the host plants.

Rice Bran Supplement Containing A Functional Substance, the Novel Peptide Leu-Arg-Ala, has Anti-Hypertensive Effects: A Double-Blind, Randomized, Placebo-Controlled Study.

Abstract
The anti-hypertensive effect of processed rice bran (PRB) was recently reported, for which the novel peptide Leu-Arg-Ala (LRA) was identified as the functional substance. The purpose of this study was to assess the anti-hypertensive effects of a rice bran supplement containing PRB in individuals with high-normal blood pressure (systolic blood pressure (SBP): 130⁻139 mmHg and/or diastolic blood pressure (DBP): 85⁻89 mmHg) or grade 1 hypertension (SBP: 140⁻159 mmHg and/or DBP: 90⁻99 mmHg). One hundred individuals with high-normal blood pressure or grade 1 hypertension were recruited to participate in this double-blind, randomized, placebo-controlled study. Participants were randomly allocated to the placebo group (n = 50) or the test group (n = 50). Each group took four test tablets (43 μg LRA/day) or four placebo tablets daily. The decrease in blood pressure in the test group compared with the placebo group was the primary outcome. Adverse events were recorded and hematological/urinary parameters measured to determine the safety of the supplement, which was the secondary outcome. In total, 87 participants completed the study. The SBP of the test group at 12 weeks was significantly lower than that of the placebo group (p = 0.0497). No serious adverse events were observed. Daily consumption of a rice bran supplement containing PRB can safely improve mildly elevated blood pressure.

Functional peptides derived from rice bran proteins.

Abstract
Rice bran has been predominantly used in the feed industry, and only recently it has attracted greater attention in terms of human nutrition with increasing knowledge of its bioactivity. A growing interest is the analysis of physiologically active peptides derived from rice bran proteins. In this paper, the bioactivities of rice bran proteins hydrolysates and peptides are reviewed based on recent studies. These enzymatic hydrolysates and peptides exert various biological activities including antioxidant, antidiabetic, anticancer and inhibitory activity for angiotensin converting enzyme (ACE), which may ultimately prevent certain chronic diseases. Nevertheless, these functionalities can be highly associated with their corresponding structural characteristics, in particular specific sequences and molecular weight distribution. This article may facilitate the expansion of the prospective applications of the bioactive peptides in a number of fields and provide some clues of the relationship between peptides structure and functionality for future research.

Effects of Farming Activities on the Biogeochemistry of Mercury in Rice-Paddy Soil Systems.

Abstract
The biogeochemistry of mercury (Hg) in rice-paddy soil systems raises concerns, given that (1) the redox potential in paddy soil favors Hg methylation and (2) rice plants have a strong ability to accumulate methylmercury (MeHg), making rice an important source for MeHg exposure to humans. Therefore, all factors affecting the behavior of Hg in rice-paddy soils might impact Hg accumulation in rice, with its subsequent potential risks. As a typical wetland, paddy soils are managed by humans and affected by anthropogenic activities, such as agronomic measures, which would impact soil properties and thus Hg biogeochemistry. In this paper, we reviewed recent advances in the effects of farming activities including water management, fertilizer application and rotation on Hg biogeochemistry, trying to elucidate the factors controlling Hg behavior and thus the ecological risks in rice-paddy soil systems. This review might provide new thoughts on Hg remediation and suggest avenues for further studies.

Developing a New Spectral Index for Detecting Cadmium-Induced Stress in Rice on a Regional Scale.

Abstract
In natural farmland ecosystems, cadmium (Cd) pollution in rice has attracted increasing attention because of its high toxicity, relative mobility, and high water solubility. This study aims to develop a spectral index for detecting Cd stress in rice on a regional scale. Three experimental sites are selected in Zhuzhou City, Hunan Province. The hyperspectral data, chlorophyll (Chl) content, leaf area index, average leaf angle, Cd concentration in soil, and Sentinel-2A images from 2017 and 2018 are collected. A new spectral index sensitive to Cd stress in rice is established based on the global sensitivity analysis of the radiative transfer model PROSPECT + SAIL (commonly called PROSAIL) model with the auxiliary of the field-measured data. The heavy metal Cd stress-sensitive spectral index (HCSI) is devised as an indicator of the degree of Cd stress in rice. Results indicate that (1) the HCSI developed based on Chl is a good indicator of rice damage caused by Cd stress, that is, low values of HCSI occur in rice subject to relatively high pollution; (2) compared with common spectral indices, such as red-edge position and red-edge Chl index, HCSI is more sensitive to Chl content with higher Pearson correlation coefficients with respect to Chl content, ranging from 0.85 to 0.95; (3) HCSI is successfully applied in Sentinel-2A images from the two different years of monitoring rice Cd stress on a regional scale. Cd stress levels in rice stabilized, and the largest area percentage of each pollution levels of Cd decreased in the following order: No pollution (i.e., 40%), low pollution (i.e., 35%), and high pollution (i.e., 25%). This study indicates that a combination of simulation data from the PROSAIL model and measured data appears to be a promising method for establishing a sensitivity spectral index to heavy metal stress, which can accurately detect regional Cd stress in crops.

Minimizing experimental artefacts in synchrotron-based X-ray analyses of Fe speciation in tissues of rice plants.

Abstract
Iron (Fe) plays an important role within environmental systems. Synchrotron-based X-ray approaches, including X-ray absorption spectroscopy (XAS), provide powerful tools for in situ analyses of Fe speciation, but beam damage during analysis may alter Fe speciation during its measurement. XAS was used to examine whether experimental conditions affect the analysis of Fe speciation in plant tissues. Even when analyzed in a cryostat at 12 K, it was found that FeIII can rapidly (within 0.5-1 min) photoreduce to FeII, although the magnitude of photoreduction varied depending upon the hydration of the sample, the coordination chemistry of the Fe, as well as other properties. For example, photoreduction of FeIII was considerably higher for aqueous standard compounds than for hydrated plant-root tissues. The use of freeze-dried samples in the cryostat (12 K) markedly reduced the magnitude of this FeIII photoreduction, and there was no evidence that the freeze-drying process itself resulted in experimental artefacts under the current experimental conditions, such as through the oxidation of FeII, although some comparatively small differences were observed when comparing spectra of hydrated and freeze-dried FeII compounds. The results of this study have demonstrated that FeIII photoreduction can occur during X-ray analysis, and provides suitable conditions to preserve Fe speciation to minimize the extent of beam damage when analyzing environmental samples. All studies utilizing XAS are encouraged to include a preliminary experiment to determine if beam damage is occurring, and, where appropriate, to take the necessary steps (such as freeze drying) to overcome these issues.

Abstract
India has the second largest number of people with type 2 diabetes (T2D) globally. Epidemiological evidence indicates that consumption of white rice is positively associated with T2D risk, while intake of brown rice is inversely associated. Thus, we explored the effect of substituting brown rice for white rice on T2D risk factors among adults in urban South India. A total of 166 overweight (BMI ≥ 23 kg/m2) adults aged 25-65 years were enrolled in a randomised cross-over trial in Chennai, India. Interventions were a parboiled brown rice or white rice regimen providing two ad libitum meals/d, 6 d/week for 3 months with a 2-week washout period. Primary outcomes were blood glucose, insulin, glycosylated Hb (HbA1c), insulin resistance (homeostasis model assessment of insulin resistance) and lipids. High-sensitivity C-reactive protein (hs-CRP) was a secondary outcome. We did not observe significant between-group differences for primary outcomes among all participants. However, a significant reduction in HbA1c was observed in the brown rice group among participants with the metabolic syndrome (-0·18 (se 0·08) %) relative to those without the metabolic syndrome (0·05 (se 0·05) %) (P-for-heterogeneity = 0·02). Improvements in HbA1c, total and LDL-cholesterol were observed in the brown rice group among participants with a BMI ≥ 25 kg/m2 compared with those with a BMI < 25 kg/m2 (P-for-heterogeneity < 0·05). We observed a smaller increase in hs-CRP in the brown (0·03 (sd 2·12) mg/l) compared with white rice group (0·63 (sd 2·35) mg/l) (P = 0·04). In conclusion, substituting brown rice for white rice showed a potential benefit on HbA1c among participants with the metabolic syndrome and an elevated BMI. A small benefit on inflammation was also observed.

[From genome analysis to construction of an integrated omics knowledgebase for crops].

Abstract
The advances in high-throughput technologies have enabled high-speed accumulation of omics data, which contain a large amount of genetic variations and their functional information. The integration and deep utilization of those data will be a long-term and difficult task, which requires highly efficient data storage and powerful data analysis and mining tools. In the past several years, our group has conducted multi-level genomic analyses in several plants, including genome assembly and annotation, comparative and population genomic studies, through collaboration with other labs inside and outside of our institution. Meanwhile, we have integrated a large amount of rice germplasm information and omics data into a structural database and developed related data query, visual display and mining web tools. Here, we summarize some of those results and discuss our next goal to construct an integrated omics knowledgebase for crops to support functional genomics and molecular design breeding.
